Description

Description: This assay employs a two-site sandwich ELISA to quantitate MPZL2 in samples. An antibody specific for MPZL2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MPZL2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MPZL2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MPZL2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: EVA1 is expressed in the thymus and in several epithelial structures early in embryogenesis. It is highly homologous to the myelin protein zero and, in thymus-derived epithelial cell lines, is poorly soluble in nonionic detergents, strongly suggesting an association to the cytoskeleton. Its capacity to mediate cell adhesion through a homophilic interaction and its selective regulation by T cell maturation might imply the participation of EVA in the earliest phases of thymus organogenesis. The protein bears a characteristic V-type domain and two potential N-glycosylation sites in the extracellular domain; a putative serine phosphorylation site for casein kinase 2 is also present in the cytoplasmic tail.
